    SpeckleNN: A unified embedding for real-time speckle pattern classification in X-ray single-particle imaging with limited labeled examples

    With X-ray free-electron lasers (XFELs), it is possible to determine the three-dimensional structure of noncrystalline nanoscale particles using X-ray single-particle imaging (SPI) techniques at room temperature. Classifying SPI scattering patterns, or "speckles", to extract single hits that are needed for real-time vetoing and three-dimensional reconstruction poses a challenge for high data rate facilities like European XFEL and LCLS-II-HE. Here, we introduce SpeckleNN, a unified embedding model for real-time speckle pattern classification with limited labeled examples that can scale linearly with dataset size. Trained with twin neural networks, SpeckleNN maps speckle patterns to a unified embedding vector space, where similarity is measured by Euclidean distance. We highlight its few-shot classification capability on new never-seen samples and its robust performance despite only tens of labels per classification category even in the presence of substantial missing detector areas. Without the need for excessive manual labeling or even a full detector image, our classification method offers a great solution for real-time high-throughput SPI experiments

    Binding of PFOS to serum albumin and DNA: insight into the molecular toxicity of perfluorochemicals

    <p>Abstract</p> <p>Background</p> <p>Health risk from exposure of perfluorochemicals (PFCs) to wildlife and human has been a subject of great interest for understanding their molecular mechanism of toxicity. Although much work has been done, the toxigenicity of PFCs remains largely unknown. In this work, the non-covalent interactions between perfluorooctane sulfonate (PFOS) and serum albumin (SA) and DNA were investigated under normal physiological conditions, aiming to elucidate the toxigenicity of PFCs.</p> <p>Results</p> <p>In equilibrium dialysis assay, the bindings of PFOS to SA correspond to the Langmuir isothermal model with two-step sequence model. The saturation binding number of PFOS was 45 per molecule of SA and 1 per three base-pairs of DNA, respectively. ITC results showed that all the interactions were spontaneous driven by entropy change. Static quenching of the fluorescence of SA was observed when interacting with PFOS, indicating PFOS bound Trp residue of SA. CD spectra of SA and DNA changed obviously in the presence of PFOS. At normal physiological conditions, 1.2 mmol/l PFOS reduces the binding ratio of Vitamin B<sub>2 </sub>to SA by more than 30%.</p> <p>Conclusion</p> <p>The ion bond, van der Waals force and hydrophobic interaction contributed to PFOS binding to peptide chain of SA and to the groove bases of DNA duplex. The non-covalent interactions of PFOS with SA and DNA alter their secondary conformations, with the physiological function of SA to transport Vitamin B<sub>2 </sub>being inhibited consequently. This work provides a useful experimental method for further studying the toxigenicity of PFCs.</p

    Preparation and biological application of antibodies against leucoanthocyanidin reductase and anthocyanidin reductase from grape berry

    Proanthocyanidins (PAs) endow wine with the flavor of bitterness and astringency. Both leucoanthocyanidin reductase (LAR) and anthocyanidin reductase (ANR) are two key enzymes of PA biosynthesis in grape berries, but the previous studies on these enzymes only focused on the transcriptional expression of these genes. Here, the full-length cDNAs of VvLAR1, VvLAR2 and VvANR, respectively, were cloned from wine grape berries and were then introduced into the pGEX-4T-1 expression vectors, which were highly expressed in Escherichia coli DH5α cells with the induction of the isopropyl-β-D-thiogalactoside (IPTG). The purified fusion proteins were used as the antigens to immunize rabbits, separately. The obtained antiserums were further purified to obtain the immunoglobulin G (Ig G) fractions, which were demonstrated to be capable of specifically immuno-recognizing the VvLAR1, VvLAR2 and VvANR from the crude protein extracts from grape berries with weight masses of approximately 43 kD. The analyses of translational expression of these enzyme genes during berry development and immunohistochemical localization of these proteins, by using the obtained antibodies, showed that a high amount of VvLAR1, VvLAR2 or VvANR was present at the pre-veraison stage and these enzyme proteins were all localized on the outer layer of the berry skin and the vascular bundle, as well as in the inner layer of the seed coat. This work provides an important basis for further studies on PA biosynthesis in grape berries.

    Insights of biosurfactant producing Serratia marcescens strain W2.3 isolated from diseased tilapia fish: a draft genome analysis

    Background Serratia marcescens is an opportunistic bacterial pathogen with broad range of host ranging from vertebrates, invertebrates and plants. S. marcescens strain W2.3 was isolated from a diseased tilapia fish and it was suspected to be the causal agent for the fish disease as virulence genes were found within its genome. In this study, for the first time, the genome sequences of S. marcescens strain W2.3 were sequenced using the Illumina MiSeq platform. Result Several virulent factors of S. marcescens such as serrawettin, a biosurfactant, has been reported to be regulated by N-acyl homoserine lactone (AHL)-based quorum sensing (QS). In our previous studies, an unusual AHL with long acyl side chain was detected from this isolate suggesting the possibility of novel virulence factors regulation. This evokes our interest in the genome of this bacterial strain and hereby we present the draft genome of S. marcescens W2.3, which carries the serrawettin production gene, swrA and the AHL-based QS transcriptional regulator gene, luxR which is an orphan luxR. Conclusion With the availability of the whole genome sequences of S. marcescens W2.3, this will pave the way for the study of the QS-mediated genes expression in this bacterium

    Factors Associated With Leisure Participation Among the Elderly Living in Long-term Care Facilities

    SummaryBackgroundA lack of participation in leisure activities often leads to depression in the elderly. This study investigated the factors impacting leisure participation among the elderly living in long-term care facilities.MethodsThis cross-sectional study recruited 309 individuals older than 65 years from six long-term care facilities located in the Taipei area. Structured in-person questionnaires were administered to assess their demographic characteristics, general self-rated health status, leisure constraints, and leisure participation.ResultsThe average frequency of leisure participation was 27.20 ± 12.48 points. The top five most popular leisure activities were watching television, walking, chatting, reading, and participating in religious activities. Elderly subjects who tended to be female, have religious beliefs, have a high school education level, be married, perceive themselves in better health, demonstrate better cognitive function and have higher scores on activities of daily living and instrumental activities of daily living scales were more likely to participate in leisure activities (p < 0.05). In contrast, those who were older, had stayed in the facility for a longer time period and had more leisure constraints were less likely to participate in leisure activities (p < 0.05). Overall, the predictive factors for leisure participation included religious beliefs, educational level, cognitive skill, instrumental activities of daily living score, depression, personal inner constraints, and structural constraints. The entire model was significant (F = 11.03, p < 0.001).ConclusionBased on the self-reported interests, health status and level of cognitive skill of elderly residents, long-term care facilities should arrange appropriate leisure activities to prevent depression and to improve quality of life

    False Data Injection Attack on Atmospheric Electric Field in Thunderstorm Warning

    Thunderstorm warning plays an important role in lightning prevention and disaster mitigation. In practical applications, thunderstorm warning system is also vulnerable to attacks, such as False Data Injection Attack (FDIA). However, there is a lack of research on False Data Injection Attack for thunderstorm warning. Therefore, this paper put forwards a FDIA method based on principal component analysis (PCA) for atmospheric electric field (AEF), which is usually used for thunderstorm warning. In the FDIA scenario, the AEF-based thunderstorm warning algorithm is also introduced with electric field differential index (EFDI). Finally, experiments are conducted based on AEF data collected by an atmospheric electric field meter (AEFM) about the real thunderstorm. The experimental results show that FDIA seriously interferes with the results of the AEF-based thunderstorm warning

    COMPARISON OF TORSO TWIST BETWEEN SLAP HIT AND ORDINARY HIT IN SOFTBALL BATTING

    Softball batters take advantage of slap hit, by positioning the batters much closer to the first base. The purpose of this study was to compare the difference of torso twist between a slap hit and an ordinary hit in softball batting. Ten female college softball batters performed slap hits and ordinary hits. Reflective markers were placed on specific landmarks for each subject and VICON motion analysis system was used to record the hits. Slap hits showed less backward rotation during the torso wind-up phase while ordinary hit showed more forward rotation during the torso follow-through phase. No difference on trunk rotation was found at impact. The findings of this study suggested that the restricted backward torso twist during the wind-up phase and the limited forward torso twist during the follow-through phase should be taken into consideration in slap hits
